Abstract

INDONESIA Tembaga (Cu) merupakan limbah yang termasuk dalam logam berat, yang berbahaya bagi organisme bahkan manusia. Mikroalga merupakan organisme uni seluler yang mampu menyerap Cu dengan baik. Konsentrasi tembaga tertentu dapat meningkatkan pertumbuhan, kadar klorofil, dan kadar lipida mikroalga. Lipida mikroalga dapat dimanfaatkan sebagai bahan baku pembuatan biodisel. Penelitian ini bertujuan untuk mengetahui pengaruh tembaga dalam bentuk CuSO4 terhadap pertumbuhan dan kadar lipida mikroalga Scenedesmes dimorphus. Metode penelitian menggunakan rancangan acak lengkap (RAL) dengan mengaplikasikan 5 perlakuan dan 5 kali ulangan, konsentrasi CuSO4, P0= 0 mg/L (kontrol) P1= 0,1 mg/L, P2= 0,2 mg/L, P3= 0,3 mg/L dan P4= 0,4 mg/L. Dikultur pada botol kultur selama 14 hari dengan menggunakan MBB (Medium Basal Bold) dan dilakukan pengamatan terhadap parameter pertumbuhan, berat kering, kadar lipida dan kadar klorofil mikroalga. Data yang di dapat kemudian di uji statistik menggunakan Analysis of Variance (ANOVA) dan diuji lanjut dengan menggunakan uji DNMRT (Duncan’s New Multiple Range Test) pada taraf 5% dan menggunakan persamaan regresi kuadratik dan persamaan regresi linear. Terdapat pengaruh yang nyata terhadap pertumbuhan, kadar lipida dan klorofil mikroalga Scenedesmus dimorphus dan tidak berpengaruh nyata terhdap berat kering mikroalga Scenedesmus dimorphus. Didapat hasil pertumbuhan tertinggi dicapai pada perlakuan CuSO4 0,3 mg/L terendah dicapai pada perlakuan CuSO4 0,4 mg/L. Berat kering tertinggi dicapai pada perlakuan CuSO4 0,1 yaitu 0,14 gram. Kadar lipida tertinggi dicapai pada perlakuan 0,3 mg yaitu 73%. Kadar klorofil a tertinggi dicapai pada perlakuan CuSO4 0,1 mg/L yaitu 10,29 mg/L dan kadar klorofil b tertinggi dicapai pada perlakuan CuSO4 0,1 mg/L yaitu 9,50 mg/L. ENGLISH Copper (Cu) is waste that is included in the heavy metal, which is harmful to the organism even humans. Microalgae are uni cellular organism that is able to absorb Cu well. Certain copper concentration could increase the growth, chlorophyll content, and lipid levels of microalgae. Microalgae lipids has potential to be biofuels. This study aimed to determine the effect of copper in the form of CuSO4 on growth and lipid content of microalgae Scenedesmes dimorphus. The research method using a complete randomized design (CRD) by applying 5 treatments and five repititions, the concentration of CuSO4, P0 = 0 mg / L (control) P1 = 0.1 mg / L, P2 = 0.2 mg / L, P3 = 0.3 mg / L and P4 = 0.4 mg / L. Cultured in a culture bottle for 14 days by using the MBB (Basal Medium Bold) then observe the growth parameters, dry weight, lipid levels and chlorophyll content of microalgae. The data is then statistically tested using Analysis of Variance (ANOVA) and tested further by using DNMRT test (Duncan's New Multiple Range Test) at 5% level and using the quadratic regression equation and the linear regression equation. The research shows there is a significant effect on growth, lipid levels and chlorophyll of microalgae Scenedesmus dimorphus, but there is not significant for dry weight of Scenedesmus dimorphus. The highest growth was obtained in the treatment of 0.3 mg / L CuSO4. The lowest was obtained in the treatment of 0.4 mg / L. The highest dry weight in the treatment CuSO4 0.1 is 0.14 grams. The highest lipid levels in the treatment of 0.3 mg is 73%. The highest levels of chlorophyll a in the treatment CuSO4 0.1 mg / L is 10.29 mg / L and the highest levels of chlorophyll b in the treatment CuSO4 0.1 mg / L is 9.50 mg / L.
