Abstract

INDONESIA Zat pengatur tumbuh berperan penting dalam produktifitas tumbuhan. Pemanfaatan zat pengatur tumbuh sintetis secara berlebihan akan berdampak negatif bagi lingkungan dan manusia. Salah satu jenis zat pengatur tumbuh yang memacu pertumbuhan tanaman adalah indole-3-acetic acid (IAA). Mikroalga memiliki potensi sebagai sumber penghasil IAA. Penelitian ini bertujuan untuk mengindentifikasi pengaruh pemberian berbagai konsentrasi filtrat pupuk kandang kotoran ayam terhadap pertumbuhan dan kadar IAA mikroalga Chlorella vulgaris. Penelitian ini merupakan penelitian eksperimental menggunakan Rancangan Acak Lengkap (RAL) dengan lima perlakuan konsentrasi filtrat pupuk kandang kotoran ayam meliputi T0 (kontrol), T1 (2,5%), T2 (5%), T3 (7,5%), dan T4 (10%). Parameter yang diukur adalah pertumbuhan C. vulgaris meliputi kerapatan sel, laju pertumbuhan, dan kadar klorofil serta kadar IAA pada mikroalga. Metode yang digunakan adalah pengujian kuantitatif dengan menggunakan spektrofotometer UV-Vis. Hasil menunjukan bahwa pemberian berbagai konsentrasi filtrat pupuk kandang kotoran ayam dapat meningkatkan pertumbuhan mikroalga C. vulgaris dibanding perlakuan kontrol. Perlakuan T3 memberikan hasil paling optimum pada semua parameter pengamatan. Kerapatan sel optimum dengan jumlah 1,163 x 107 sel/mL. Rata-rata laju pertumbuhan optimum dengan jumlah 0,845 pembelahan sel/hari. Kadar klorofil-a dan klorofil-b optimum masing-masing dengan jumlah 13,968 mg/L dan 4,202 mg/L. Nilai optimum kadar IAA yang diperoleh dengan jumlah 0,417 ppm. ENGLISH Plant growth regulators play an important role in the productivity of a plant. Utilization of synthetic growth regulator substances in excess that have a negatively impact on the environment and humans. One type of growth regulator substances that stimulate plant growth is indole-3-acetic acid (IAA). Microalgae have the potential as a source of IAA. This study aimed to indentify influence of varying concentrations filtrate from manure on growth and amount of IAA in Chlorella vulgaris. This research was an experimental study using a Completely Randomized Design (CRD) with five treatments the concentration of the filtrate from chicken manure, i.e. T0 (control), T1 (2.5%), T2 (5%), T3 (7.5%), and T4 (10%). Parameters measured were the growth of Chlorella vulgaris include cell density, growth rate, chlorophyll content and amount of IAA in the microalgae. The method used is quantitative testing by using a spectrophotometer UV-Vis. The results showed that various concentrations of filtrate from the manure can affect the growth of C. vulgaris more than control treatment. The T3 treatment showed the optimum results for all thorough parameters. The optimum cell density with a value of 1,163 x 107 cells / mL. The optimum average of growth rate with a value of 0,845 cell division / day. The optimum levels of chlorophyll-a and chlorophyll-b apart with a value of 13,968 mg / L and 4,202 mg / L. The optimum amount of IAA with a value of 0,417 ppm.
