Abstract

In vitro culture through subculture is a solution propagation technique in fulfilling the market demand for chrysanthemum plants which have the 3rd largest export value in ASEAN. The combination of foliar fertilizer with BAP hormone can be used as an alternative media substitute with complete nutrients and affordable prices and is able to support the success of subculture in shoot multiplication. The purpose of this study was to determine the most optimal foliar fertilizer media and BAP concentration on the growth results of subculture of Pasopati variety chrysanthemum plants. This experimental study used a non-factorial Completely Randomized Design (CRD) method and descriptive methods presented in the form of graphs and tables for 10 combination treatments and 4 replications, which include: P0 = ½ MS + 1 mg L-1 BAP (control); P1 = G1 + 0.5 mg L-1 BAP; P2 = G1 + 1 mg L-1 BAP; P3 = G1 + 1.5 mg L-1 BAP; P4 = G2 + 0.5 mg L-1 BAP; P5 = G2 + 1 mg L-1 BAP; P6 = G2 + 1.5 mg L-1 BAP; P7 = G3 + 0.5 mg L-1 BAP; P8 = G3 + 1 mg L-1 BAP; P9 = G3 + 1.5 mg L-1 BAP. The findings indicated that the application of foliar fertilizer and BAP concentration had a significant positive effect on the growth performance during the subculture stage of the Pasopati chrysanthemum variety under in vitro conditions. The most effective treatment was identified as P2 (medium supplemented with foliar fertilizer 32:10:10) combined with 1 mg L-1 BAP, which resulted in the earliest shoot emergence at an average of 3 days after initiation (DAI), an average shoot height of 0.63 cm, and the formation of green spots (shoot primordia) on the callus morphology. This treatment combination demonstrates potential as an alternative medium for the subculture of the Pasopati chrysanthemum variety.
