Abstract

INDONESIA: Daun stevia memiliki gizi yang tinggi dan bermanfaat bagi kesehatan tubuh karena stevia mengandung protein, lemak, serat dan karbohidrat. Perbanyakan secara in vitro inisiasi eksplan nodus stevia dengan penambahan media dasar pupuk daun dan zat pengatur tumbuh alami air kelapa menjadi solusi untuk mengatasi permasalahan ini. Tujuan dari penelitian ini yaitu untuk mengetahui kombinasi media dasar pupuk daun 32-10-10 dan ZPT alami air kelapa berpengaruh terhadap hasil inisiasi planlet nodus stevia. Metode yang digunakan dalam penelitian ini adalah Rancangan Acak Lengkap (RAL) dengan 5 perlakuan dan 4 ulangan yaitu M0= Pupuk Daun 2 g L-1, M1 = Pupuk Daun 2 g L-1 + air kelapa 200 ml L-1, M2 = Pupuk Daun 2 g L-1 + air kelapa 250 ml L-1, M3 = Pupuk Daun 2 g L-1 + air kelapa 300 ml L-1, M4 = Pupuk Daun 2 g L-1 + air kelapa 350 ml L-1. Hasil penelitian menunjukan bahwa perlakuan dengan penambahan Pupuk Daun 2 g L-1 air kelapa 350 ml L-1 menghasilkan jumlah tunas terbanyak yaitu 11 tunas yang muncul, memberikan hasil tertinggi pada jumlah daun dengan rata-rata 48,3 dan eksplan tertinggi yaitu 8,0 serta jumlah akar 3,25. Tingkat hidup eksplan mencapai 100% menunjukan Tingkat keberhasilan kultur jaringan yang cukup tinggi. Penambahan konsentrasi air kelapa pada media kultur menunjukkan peningkatan signifikan dalam jumlah tunas, jumlah daun, tinggi eksplan dan jumlah akar. Hasil ini mengindikasikan bahwa air kelapa berpotensi sebagai suplemen alami yang efektif dalam meningkatkan kualitas pertumbuhan stevia secara in vitro. ENGLISH: Stevia leaves have high nutrition and are beneficial for body health because stevia contains protein, fat and carbohydrates and minerals. In vitro propagation of stevia node explant initiation with the addition of leaf fertilizer base medium and coconut water natural growth regulator is a solution to overcome this problem. The purpose of this study was to determine the combination of leaf fertilizer base media and coconut water natural ZPT effect on the results of stevia node planlet initiation. The method used in this study is a completely randomized design with 5 treatments and 4 replicates, namely M0 = Leaf Fertilizer 2 g L-1, M1 = Leaf Fertilizer 2 g L-1 + coconut water 200 ml L-1, M2 = Leaf Fertilizer 2 g L-1 + coconut water 250 ml L-1, M3 = Leaf Fertilizer 2 g L-1 + coconut water 300 ml L-1, M4 = Leaf Fertilizer 2 g L-1 + 350 ml L-1 coconut water. The results showed that the treatment with the addition of leaf fertilizer 2 g L-1 coconut water 350 ml L-1 produced the highest number of buds, namely 11 buds that appeared, giving the highest results in the number of leaves with an average of 48.3 and the highest explants, namely 8.0 and the number of roots 3.25. The explant survival rate reached 100%, indicating a fairly high success rate of tissue culture. The addition of coconut water concentration to the culture medium showed a significant increase in the number of shoots, number of leaves, explant height and number of roots. These results indicate that coconut water has the potential as an effective natural supplement in improving the quality of stevia growth in vitro.
