Abstract

INDONESIA Tanaman kemang merupakan tanaman buah yang terancam keberadaannya karena populasinya yang terus menurun. Penelitian ini bertujuan untuk mendapatkan kondisi pertumbuhan kemang dalam kultur jaringan serta menganalisa pengaruh media WPM (woody plant medium) dengan penambahan zat pengatur tumbuh (ZPT) alami air kelapa muda dan tua terhadap pertumbuhan kemang. Untuk mengoptimalkan pemanfaatan ZPT tersebut perlu didukung dengan proses sterilisasi media dan eksplan yang efektif. Percobaan dilaksanakan di Laboratorium Kultur Jaringan Jurusan Agroteknologi Fakultas Sains dan Teknologi UIN Sunan Gunung Djati Bandung dari Mei-Juli 2013. Terdapat tiga tahap percobaan. Pertama, proses sterilisasi eksplan dengan tiga perlakuan yaitu perbedaan kosentrasi dan lama waktu aplikasi larutan sterilan yang dilakukan sebanyak dua ulangan. Kedua, pengujian air kelapa sebagai hormon untuk memacu proses organogenesis langsung eksplan daun tanaman kemang. Ketiga adalah perbandingan kecepatan pertumbuhan antara kemang, krisan dan anggrek. Hasil penelitian menunjukkan bahwa dalam penelitian ini tidak terdapat kombinasi konsentrasi yang optimum antara perlakuan konsentrasi air kelapa muda dan air kelapa tua terhadap perbanyakan tanaman buah kemang. Namun terdapat respon pertumbuhan kalus pada perlakuan WPM + 0,1 ppm IAA dan WPM + 5% kelapa muda (KM) dengan awal muncul kalus pada umur 4 MST. ENGLISH Kemang is one of many tree fruit plant in Indonesia that in rare condition becahuge threated deforestation. This study aims to obtain Kemang growth conditions in tissue culture as well as to analyze the influence of the media WPM (woody plant medium) with the addition of plant growth regulators naturally young and ouse of ld coconut water on the growth of Kemang. To optimize the use of these plant hormone need supported by the media and explant sterilization process is effective. The research conducted at the Laboratory of Tissue Culture Agrotechnology State Islamic University Sunan Gunung Djati Bandung, from March to July 2013. There are three stages of the experiment. First, sterilization process of explants with three treatments was a time and the concentration difference sterilant applications with two replications. Second, testing of young and old coconut water as a hormone to stimulate the process of direct organogenesis kemang plant leaf explants. Third was the growth rate of kemang, chrysanthemums and orchids. The results showed that there was no effect of concentrate young and old coconut water on kemang propagation, but using WPM + IAA 0,1 ppm and WPM + 5% young coconut water (KM) produced callus at 4 WAP.
