Abstract

INDONESIA Fosfat merupakan nutrien esensial yang diperlukan oleh tanaman dalam pertumbuhan dan perkembangannya. Namun ketersediaan fosfat terlarut di dalam tanah jumlahnya masih rendah. Aktivitas bakteri pelarut fosfat dapat dimanfaatkan untuk penyediaan unsur hara fosfat bagi tanaman. Bakteri pelarut fosfat mampu mengeluarkan asam-asam organik dan enzim fosfatase. Bakteri pelarut fosfat sering ditemukan berasosiasi di dalam tanah rizosfer. Mikroba rizosfer dapat dimanfaatkan sebagai pupuk hayati bagi tanaman, salah satunya bagi tanaman cabai merah. Cabai merah merupakan jenis tanaman hortikultura, buahnya mengandung zat gizi dan banyak digunakan untuk keperluan industri makanan dan obat-obatan. Penelitian ini bertujuan untuk mengidentifikasi aktivitas fosfatase bakteri pelarut fosfat isolat K2.BR.7 pada beberapa variasi konsentrasi fosfat, mengidentifikasi kurva pertumbuhan dari isolat K2.BR.7, dan menganalisis pengaruh aplikasi bakteri pelarut fosfat isolat K2.BR.7 terhadap perkecambahan tanaman cabai merah. Skrining bakteri pelarut fosfat menggunakan metode titik, kurva pertumbuhan dilakukan dengan mengukur tingkat kekeruhan OD (Optical Density) menggunakan spektrofotometer pada panjang gelombang () 600 nm, dan pengaruh aplikasi bakteri pelarut fosfat dengan mengamati parameter tinggi tanaman, panjang akar, dan berat basah. Data hasil aplikasi dianalisis secara statistik dengan metode analisis variansi satu arah (one way ANOVA) pada taraf signifikansi 5% dan uji lanjut Duncan. Isolat K2.BR.7 berpengaruh secara nyata dalam membentuk zona bening pada konsentrasi pikovskaya 6,67% dengan rata-rata lebar zona bening 1,82 cm. Kurva pertumbuhan isolat K2.BR.7 menunjukkan pertumbuhan diauxic dengan 3 fase eksponensial. Aplikasi bakteri pelarut fosfat isolat K2.BR.7 pada perlakuan P1B2, P1B6, dan P1B8 berpengaruh secara nyata terhadap tinggi tanaman cabai merah. ENGLISH Phosphate is an essential nutrient required by plants in growth and development. But the availability of phosphate dissolved in the amount of land is still low. The activity of phosphate solubilizing bacterial can be used for the supply of phosphate nutrients for plants. Phosphate solubilizing bacterial capable of removing organic acids enzymes phosphatase. Phosphate solubilizing bacterial often found associated in the rhizosphere soil. Rhizosphere microbes can be used as a biological fertilizer for plants, one of which is a red chili plant. Red chili is a horticultural, the fruit contains nutrients and is widely used for the food industry and medicine. This study aims to identify the phosphatase activity of phosphate solubilizing bacterial isolates K2.BR.7 on some variation of phosphate concentrations, identifies the growth curve of the isolates K2.BR.7, and analyze the effect of the application of phosphate solubilizing bacterial on red chili germination. Phosphate solubilizing bacterial screening methods point, the growth curve is done by measuring the turbidity OD (Optical density) using a spectrophotometer at a wavelength of () 600 nm, and the influence of phosphate solubilizing bacterial applications by observing parameters plant height, root length, and fresh weight. Data application results were statistically analyzed by the methode of one-way analysis of variance (one way ANOVA) at the 5% significance level and further test Duncan. Isolates K2.BR.7 significant influence in shaping the halozone on pikovskaya concentration of 6,67% with an average of 1,82 cm wide halozone. Isolates K2.BR.7 showed growth curve diauxic with 3 exponential phase. Application of phosphate solubilizing bacterial isolates K2.BR.7 on treatment P1B2, P1B6, and P1B8 significantly affect plant height red chili.
