Abstract

INDONESIA α-amilase merupakan enzim ekstraseluler yang mampu memutuskan ikatan 1,4-αD-glikosidik antar monomer glukosa pada rantai linier amilosa. Salah satu bakteri penghasil α-amilase adalah Bacillus sp. K2Br5. Bacillus sp. K2Br5 merupakan bakteri yang berbentuk tak beraturan, berwarna putih, tepian tak beraturan, elevasi datar, dan permukaan mengkilat. Penelitian ini dilakukan untuk mengidentifikasi pengaruh penambahan variasi ion logam, variasi konsentrasi ZnSO4 dan agen pengkhelat EDTA terhadap aktivitas α-amilase dari Bacillus sp. K2Br5. Pegujian aktivitas α-amilase dilakukan menggunakan metode DNS, sedangkan untuk penentuan kadar protein total dilakukan dengan metode Bradford. Aktivitas spesifik fraksi 60% α-amilase sebesar 0,3779 U/mg. Ketika dilakukan penambahan ion logam Zn2+ dan Ca2+ aktivitas spesifik α-amilase meningkat dengan aktivitas spesifik masing-masing sebesar 0,6428 U/mg dan 0,5178 U/mg, dan menurun setelah penambahan ion logam Cu2+, Na+, dan Mn2+ dengan masingmasing aktivitas sebesar 0,3362 U/mg, 0,2444 U/mg, dan 0,1976 U/mg. Penambahan variasi konsentrasi ZnSO4 menunjukan bahwa semakin besar konsentrasi ZnSO4 maka semakin berkurang aktivitas spesifik α-amilase. Penambahan agen pengkhelat EDTA dapat menurunkan aktivitas spesifik αamilase, semakin bertambahnya konsentrasi EDTA maka aktivitas spesifik αamilase semakin mengalami penurunan. ENGLISH α-amylase is an extracellular enzyme that is able to break the 1,4-α-D-glycosidic link between glucose monomers in the amylose linear chain. One of the α-amylase producing bacteria is Bacillus sp. K2Br5. Bacillus sp. K2Br5 is an irregularly shaped, white bacteria, irregular edges, flat elevation, and shiny surface. This research was conducted to find information about the effect of metal ions, the concentration of ZnSO4 and chelating agent EDTA on α-amylase activity of Bacillus sp. K2Br5. Tests carried out were α-amylase activity test and determination of total protein content. Tests for α-amylase activity were carried out using the DNS method, while for total protein test carried out by Bradford method. GunakThe specific activity of the 60% α-amylase fraction was 0.3779 U/mg. When adding metal ions Zn2+ and Ca2+ specific activity α-amylase increases with activity of 0.6428 U/mg and 0.5178 U/mg, and decreases after adding metal ions Cu2+, Na+, and Mn2+ with each activity . 0.3362 U/mg, 0.2444 U/mg, and 0.1976 U/mg. The determination of ZnSO4 concentration showed that the greater the concentration of ZnSO4, the smaller the α-amylase activity. Addition of EDTA chelating agents can reduce α-amylase specific activity, the greater the concentration of EDTA, the smaller the specific activity of α-amylase.
