Abstract

INDONESIA Salah satu faktor yang berpengaruh terhadap kanker payudara adalah estrogen dan reseptor estrogen alfa. Estrogen yang berikatan dengan reseptor estrogen mengakibatkan proliferasi sel secara abnormal. Inhibisi reseptor estrogen menjadi pendekatan utama dalam pencegahan dan terapi kanker payudara. Senyawa daidzein diketahui sebagai inhibitor yang dapat berikatan dengan reseptor estrogen. Daidzein ini akan berkompetisi dengan estrogen. Penelitian dilakukan untuk mengetahui kemampuan inhibisi daidzein terhadap reseptor estrogen alfa menggunakan pendekatan in silico. Pendekatan in silico menggunakan metode penambat molekul untuk memprediksi interaksi dan energi bebas antara ligan dan reseptor. Penelitian ini dilakukan di Laboratorium Data Processing Jurusan Biologi Fakultas Sains dan Teknologi, Universitas Islam Negeri Sunan Gunung Djati Bandung pada bulan Desember 2017 sampai Februari 2018. Hasil analisis menunjukkan bahwa senyawa daidzein sebagai inhibitor yang memiliki potensi estrogenik berdasarkan nilai binding affinity (-9,4 kkal/mol) lebih kuat dibanding kontrol negatif (17-β Estradiol) (-8,9 kkal/mol) namun masih lebih lemah dibanding kontrol positif (Dihydrobenzoxanthiin) (-11,3 kkal/mol). Semakin rendah nilai binding affinity menunjukkan kemampuan pengikatan yang tinggi antara daidzein dan reseptor. Kesimpulan dari penelitian ini daidzein merupakan inhibitor bagi reseptor estrogen alfa secara in silico. ENGLISH One of the factors that affect breast cancer is estrogen and estrogen receptor alpha. Estrogens binding to estrogen receptors caused abnormal cell proliferation. Estrogen receptor inhibition becomes a major approach in the prevention and treatment of breast cancer. Daidzein compounds are known as inhibitors that could bind to estrogen receptors. Daidzein will competing with endogenous estrogen. The study was conducted to determine the ability of daidzein inhibition to estrogen receptors alpha using the in silico study. In silico study uses molecular docking methods to predict the interaction and free energy between ligand and receptor. This research was conducted at Data Processing Laboratory of Department Biology Faculty of Science and Technology, State Islamic University of Sunan Gunung Djati Bandung in December 2017 until February 2018. The result showed that daidzein compound as inhibitor having estrogenic potency based on binding affinity value (-9,4 kcal / mol) stronger than negative control (17-β Estradiol) (-8.9 kcal/mol) but still weaker than positive control (Dihydrobenzoxanthiin) (-11.3 kcal/mol). The lower the binding affinity value shows the high binding ability between daidzein and receptor. In conclusion, daidzein is an inhibitor for estrogen receptors alpha on study in silico.
