Abstract

INDONESIA Penyakit Parkinson merupakan penyakit neurodegeneratif terbanyak kedua setelah penyakit Alzheimer yang terjadi akibat degenerasi pada otak utama di bagian substansia nigra. Proses neuroprotektif dari senyawa antioksidan dapat menghambat dan mencegah terjadinya gejala penyakit Parkinson. Sumber senyawa antioksidan alami yaitu microgreens kemangi (Ocimum basilicum L.). Tujuan dari penelitian ini yaitu untuk mengetahui pengaruh ekstrak microgreens kemangi terhadap kelulusan hidup, gerak lokomotor, kadar malondialdehid (MDA) dan kadar dopamin dari penyakit Parkinson pada lalat buah (Drosophila melanogaster) yang diinduksi paraquat serta untuk mengetahui aktivitas antioksidan dan kandungan senyawa polifenol total, klorofil, dan karotenoid pada ekstrak microgreens kemangi. Penelitian ini merupakan penelitian eksperimental menggunakan rancangan acak lengkap dengan empat perlakuan dan lima ulangan. Hasil dari pengujian aktivitas antioksidan ekstrak microgreens kemangi didapatkan IC50 sebesar 49,883 µg/mL yang bersifat sangat kuat dan kadar polifenol total sebesar 190 mg GAE/g ekstrak, kadar klorofil total sebesar 19,17 mg/g serta kadar karotenoid sebesar 13,82 µmol/g. Berdasarkan hasil pengujian kelulusan hidup, bahwa lalat buah perlakuan ekstrak microgreens kemangi mampu menghasilkan nilai tertinggi sebesar 54,66%, sedangkan terendah pada perlakuan paraquat sebesar 37,99%. Gerak lokomotor diukur dari uji geotaksis negatif, bahwa lalat buah perlakuan ekstrak microgreens mampu menghasilkan nilai tertinggi sebesar 60%, sedangkan terendah pada perlakuan paraquat sebesar 40%. Adapun kadar malondialdehid tertinggi diperoleh pada lalat buah perlakuan paraquat sebesar 21,81 nMol/mL, sedangkan terendah pada perlakuan ekstrak microgreens kemangi sebesar 19,72 nMol/mL. Kemudian, data menunjukkan perlakuan ekstrak microgreens kemangi juga mampu menghasilkan kadar dopamin tertinggi sebesar 0,13, dan perlakuan paraquat menghasilkan kadar dopamin terendah sebesar 0,03. Kesimpulan dari penelitian ini yaitu ekstrak microgreens kemangi dapat berperan sebagai neuroprotektif terhadap penyakit Parkinson yang diinduksi paraquat pada lalat buah. ENGLISH Parkinson's disease is the second most neurodegenerative disease after Alzheimer's disease which occurs due to degeneration in the main brain in the substantia nigra. The neuroprotective process of antioxidant compounds can inhibit and prevent the occurrence of symptoms of Parkinson's disease. The source of natural antioxidant compounds is basil microgreens (Ocimum basilicum L.). The purpose of this study was to determine the effect of basil microgreens extract on survival rate, locomotor motion, malondialdehyde (MDA) and dopamine levels of Parkinson's disease in fruit flies (Drosophila melanogaster) which were induced by paraquat and to determine antioxidant activity and total polyphenol compounds, chlorophyll, and carotenoids in extract of basil microgreens. This study was an experimental study using a completely randomized design with four treatments and five replications. The results of testing the antioxidant activity of extract of basil microgreens obtained IC50 of 49.883 µg/mL which was very strong and total polyphenol levels of 190 mg GAE/g extract, total chlorophyll levels of 19.17 mg/g and carotenoid levels of 13.82 µmol/g. Based on the results of the survival rate test, fruit fly treated by extract of basil microgreens was able to produce the highest value of 54.66%, while the lowest result is go for paraquat treatment 37.99%. Locomotor motion was measured from the negative geotaxis test, that the fruit fly treated by extract of microgreens was able to produce the highest value of 60%, while the lowest value goes for paraquat treatment 40%. The highest levels of malondialdehyde were obtained in paraquat fruit flies at 21.81 nMol/mL, while the lowest in basil extracts of microgreens was 19.72 nMol/mL. Then, the data showed the treatment of extract of basil microgreens was also able to produce the highest dopamine levels of 0.13, and paraquat treatment produced the lowest dopamine levels of 0.03. The conclusion of this study is that extract of basil microgreens can act as a neuroprotective against Parkinson's disease induced by paraquat in fruit flies.
