Abstract
The determination of low alcohol content is very important to determine the halalness of a drink. The currently commonly used method of analyzing alcohol content is gas chromatography (GC), but this method has several drawbacks, namely high operating costs, not all laboratories can access it and only one sample per runtime. Therefore, it is proposed a method which is more economical, simple, quantitative and non-chromatographic in determining alcohol content using the validated spectrophotometric UV-Vis principle of microdifusion and is applied in samples of low alcoholic beverages. This method is based on the oxidation of ethanol compounds using potassium permanganate under alkaline conditions and was detected by spectrophotometry UV-Vis, optimized at different temperatures (35℃.45℃.55℃) and different times (20 to 60 minutes). With a sample analysis time of 50 minutes at a temperature of 35℃, it has good validity of 0.9984, the coefficient of variation of 0.122% shows good accuracy, 95.28% percent recovery shows good accuracy, detection limits (LOD) and quantization limits. (LOQ) of 0.00932 and 0.01%, respectively. This method has been applied successfully for the determination of alcohol content in beverage samples with low alcohol content. The measurements obtained by the microdifusion method using spectrophotometry UV-Vis were in accordance with the alcohol content listed on the package, which was 0.076%. This method is expected to be a routine analysis in determining alcohol content in laboratory and industrial samples.
