Abstract

Bakteri Esherichia coli merupakan bakteri yang hidup di saluran pencernaan. Bakteri ini dapat menjadi patogen dan mengeluarkan racun berupa enterotoksin yang menyebabkan penyakit diare juga penyakit lain seperti infeksi saluran kemih, gasteroentritis atau radang saluran pencernaan hingga dapat menyebabkan meningitis. Ekstrak daun mangga arumanis (Mangifera indica L) mengandung senyawa metabolit sekunder yang aktif sebagai antibakteri seperti flavonoid, tanin, alkaloid dan saponin. Penelitian ini bertujuan untuk mengidentifikasi senyawa metabolit sekunder yang terkandung dalam ekstrak daun mangga arumanis dan untuk mengetahui daya hambat Ekstrak daun mangga arumanis (Mangifera indica L) terhadap bakteri Esherichia coli. Penelitian ini menggunakan metode maserasi bertingkat dalam ekstraksi daun mangga arumanis dengan pelarut n-heksana , etil asetat dan etanol. Setiap ekstrak diuji aktivitas antibakteri dengan metode difusi cakram kertas (disk diffusion). Hasil penelitian menunjukan bahwa pada ekstrak n-heksana tidak terbentuk zona hambat, pada ekstrak etil asetat terbentuk zona hambat sebesar 2,49 mm pada konsentrasi 250 mg/ml, 1,1 mm pada konsentrasi 150 mg/ml dan 0,55 mm pada konsentrasi 100 mg/ml, pada ekstrak etanol terbentuk zona hambat sebesar 1,1 mg/ml pada konsentrasi 100 mg/ml, 1,57 mm pada konsentrasi 150 mg/ml dan 2,87 mm pada konsentrasi 250 mg/ml. Hasil yang diperoleh menunjukan bahwa ekstrak n-heksana, etil asetat dan etanol daun mangga arumanis memiliki aktivitas antibakteri yang lemah karena zona hambat yang terbentuk kurang dari 5 mm. Esherichia coli bacteria are bacteria that live in the digestive tract. These bacteria can become pathogens and release toxins in the form of enterotoxins that cause diarrhea as well as other diseases such as urinary tract infections, gastroenteritis or inflammation of the digestive tract which can cause meningitis. The leaf extract of mango arumanis (Mangifera indica L) contains secondary metabolites which are active as antibacterial compounds, such as flavonoids, tannins, alkaloids and saponins. This study aims to identify secondary metabolites contained in the arumanis mango leaf extract and to determine the inhibition power of arumanis mango leaf extract (Mangifera indica L) against Esherichia coli bacteria. This study used a multilevel maceration method in the extraction of arumanis mango leaves with n-hexane, ethyl acetate and ethanol as solvents. Each extract was tested for antibacterial activity by the paper disc diffusion method (disk diffusion). The test results showed that the n-hexane extract did not form an inhibition zone, the ethyl acetate extract formed an inhibition zone of 2.49 mm at a concentration of 250 mg / mL, 1.1 mm at a concentration of 150 mg / mL and 0.55 mm at a concentration of 100 mg / mL, at The ethanol extract formed an inhibition zone of 1.1 mg / mL at a concentration of 100 mg / mL, 1.57 mm at a concentration of 150 mg / mL and 2.87 mm at a concentration of 250 mg / mL. The results obtained showed that the extracts of n-hexane, ethyl acetate and ethanol from arumanis mango leaves had weak antibacterial activity because the inhibition zone formed was less than 5 mm.
