Abstract

INDONESIA : Karuk (Piper sarmentosum) adalah tanaman semak yang termasuk ke dalam famili Piperceae yang biasa digunakan sebagai obat tradisional untuk mengatasi berbagai penyakit seperti batuk dan asma. Daun karuk mengandung salah satu senyawa metabolit sekunder yaitu flavonoid yang berpotensi sebagai antioksidan. Penelitian ini bertujuan untuk mengidentifikasi kandungan senyawa metabolit sekunder dalam daun karuk, mengidentifikasi total flavonoid, serta aktivitas antioksidan di dalam daun karuk. Ekstraksi daun karuk digunakan dengan metode maserasi menggunakan pelarut etanol dan etil asetat. Analisis kualitatif daun karuk diuji dengan reaksi warna terhadap senyawa alkaloid, flavonoid, tanin, saponin, dan steroid/terpenoid. Sedangkan analisis kuantitatif daun karuk dilakukan secara spektrofotometri UV-Vis yaitu uji total flavonoid dengan AlCl3 pada panjang gelombang 435 nm dan uji aktivitas antioksidan menggunakan metode DPPH (1,1-diphenyl-2-picrylhydrazyl) pada panjang gelombang 517 nm. Dari penelitian ini diperoleh hasil bahwa ekstrak etanol daun karuk positif mengandung senyawa golongan alkaloid, flavonoid, tanin, saponin, dan steroid, sedangkan ekstrak etil asetat daun karuk positif mengandung senyawa golongan alkaloid, flavonoid, tanin, dan steroid. Total flavonoid ekstrak etanol daun karuk sebesar 66,13 mgQE/g, sedangkan ekstrak etil asetat sebesar 57,92 mgQE/g. Ekstrak etanol daun karuk dengan nilai IC50 sebesar 122,07 μg/mL, sedangkan ekstrak etil asetat sebesar 110,05 μg/mL dengan asam askorbat sebagai pembanding sebesar 32,03 μg/mL. Hasil penelitian ini menunjukkan daun karuk memiliki potensi aktivitas antioksidan yang sedang. ENGLISH : Karuk (Piper sarmentosum) is a shrub that belongs to the Piperceae family which is commonly used as a traditional medicine to treat various diseases such as cough and asthma. Karuk leaves contain one of the secondary metabolite compounds, namely flavonoids which have the potential as antioxidants. This study aims to identify the content of secondary metabolites in karuk leaves, identify total flavonoids, and antioxidant activity in karuk leaves. Karuk leaf extraction was used by maceration method using ethanol and ethyl acetate as solvents. Qualitative analysis of karuk leaves was tested by color reaction to alkaloids, flavonoids, tannins, saponins, and steroids/terpenoids. While the quantitative analysis of karuk leaves was carried out using UV-Vis spectrophotometry, namely the total flavonoid test with AlCl3 at a wavelength of 435 nm and the antioxidant activity test using the DPPH method (1,1-diphenyl-2-picrylhydrazyl) at a wavelength of 517 nm. From this research, it was found that ethanol extract of karuk leaves positively contains alkaloids, flavonoids, tannins, saponins, and steroid, while the ethyl acetate extract positively contains alkaloids, flavonoids, tannins, and steroid. The total flavonoids of the ethanol extract of karuk leaves were 66,13 mgQE/g, while the ethyl acetate extract was 57,92 mgQE/g. The ethanol extract of karuk leaves with IC50 value of 122,07 μg/mL, while the ethyl acetate extract of 110.05 μg/mL with ascorbic acid as a comparison of 32.03 μg/mL. The results of this study indicate that karuk leaves have moderate antioxidant activity potential.
