Abstract
The propagation of the Arshanti Agrihorti chrysanthemum variety is generally carried out using tissue culture techniques. This technique requires the right media to ensure optimal seedling growth. In accordance with the objective of this research, which is to find alternative media that can optimize the development of Arshanti Agrihorti chrysanthemum variety. This research compares MS (full and ½ concentration) and growmore leaf fertilizer (2 and 3 g L⁻¹) with the substitution of TDZ at three levels (0, 0,5, and 1 ppm), resulting in a total of 12 treatment combinations. This research uses a descriptive method with two types of data, qualitative and quantitative. Observations were made by monitoring the emergence of shoots, shoot height, number of leaves, and other responses from the cultured explants. The results of the research over 9 MSI indicate that a concentration of MS + 0,5 ppm TDZ can accelerate the induction of chrysanthemum shoots and is beneficial for the propagation of leaves and shoots. The leaf fertilizer medium at 3 g L-1 without TDZ can induce roots more optimally compared to the MS medium. Meanwhile, for the leaf color category, the ½ MS medium without TDZ and the leaf fertilizer at 3 g L-1 are considered good media. This research shows that the use of MS media with the addition of TDZ (0,5 and 1 ppm) is effective for inducing the growth of shoots from explants of the Arshanti Agrihorti chrysanthemum variety. Therefore, further development is needed regarding the utilization of TDZ in chrysanthemum subcultures.
