Abstract
INDONESIA : Diabetes melitus merupakan penyakit gangguan metabolisme yang ditandai dengan hiperglikemia akibat dari penurunan sekresi insulin, atau resistensi insulin. Diabetes melitus ada beberapa jenis yaitu, DM tipe 1, DM tipe 2, dan diabetes gestasional. Persentase DM tipe 2 lebih tinggi dibandingkan jenis lainnya, yakni 90-95% pada tahun 2013. Produksi glukosa dari jalur glikogenolisis memberikan pengaruh terhadap penderita diabetes saat kadar gula darah tinggi, dalam hal ini terlibat Human Liver Glycogen Phophorylase (HLGP). Senyawa dalam daun yakon (Smallanthus sonchifolius) yaitu Enhydrin, Sonchifolin, dan Uvedalin diduga optimal dalam pengobatan DM tipe 2, maka dari itu perlu dilakukan pengujian In silico yang menggunakan kemampuan komputer dalam proses mendesain obat, sebagai komplemen dari in vitro dan in vivo. Tujuan penelitian ini adalah untuk mengetahui nilai binding affinity dalam senyawa yang terdapat dalam daun yakon (Smallanthus sonchifolius) dan senyawa manakah yang paling efektif dalam pengobatan DM tipe 2. Penelitian ini dilakukan di Laboratorium Pengolahan Data, Jurusan Biologi, Fakultas Sains dan Teknologi, Universitas Islam Negeri Sunan Gunung Djati Bandung. Kegiatan penelitian dilaksanakan pada Desember 2019 sampai dengan Februari 2020. Hasil analisis menunjukkan nilai binding affinity ketiga senyawa yaitu, Enhydrin -7,1 kcal/mol, Sonchifolin -7,7 kcal/mol, dan Uvedalin -8,3 kcal/mol. Dari ketiga senyawa tersebut, Uvedalin merupakan senyawa paling efektif dalam pengobatan DM tipe 2 karena memiliki nilai binding affinity terendah yaitu -8,3 kcal/mol. ENGLISH : Diabetes mellitus is a metabolic disorder characterized by hyperglycemia resulting from decreased insulin secretion, or insulin resistance. There are several types of diabetes mellitus, such as T1DM, T2DM, and gestational diabetes. The percentage of T2DM is higher than other types, which is 90-95% in 2013. The glycogenolysis pathway plays a role in diabetics when high blood sugar levels are involved, in this case Human Liver Glycogen Phophorylase (HLGP). Compounds in yacon leaf (Smallanthus sonchifolius) such as Enhydrin, Sonchifolin, and Uvedalin are reported to be optimal in the treatment of T2DM, therefore it is necessary to do an in silico test that uses the ability of computers in the process of designing drugs, as a complement of in vitro and in vivo. The purpose of this study was to determine the value of binding affinity in compounds contained in yacon leaf (Smallanthus sonchifolius) and which compound is most effective in the treatment of T2DM. This research was conducted at Data Processing Laboratory, Department of Biology, Faculty of Science and Technology, State Islamic University of Sunan Gunung Djati Bandung. The research activities were carried out in December 2019 to February 2020. The results of the analysis showed the binding affinity of the three compounds are Enhydrin -7.1 kcal/mol, Sonchifolin -7.7 kcal/mol, and Uvedalin -8.3 kcal/mol. Uvedalin is the most effective compound in the treatment of T2DM because it has the lowest binding affinity value -8.3 kcal/mol
