Abstract

INDONESIA : Kalsium oksalat merupakan komponen utama dalam batu ginjal yang terbentuk karena terjadinya supersaturasi pada urine. Metode pengobatan dan pencegahan penyakit batu ginjal secara medis maupun tradisional masih belum memberikan hasil yang efektif, sehingga upaya pencarian tumbuhan yang berpotensi untuk mencegah pembentukan batu ginjal terus dilakukan. Tumbuhan Aglaia elaeagnoidea, khususnya bagian batangnya, diketahui memiliki kandungan fenolik yang tinggi dimana senyawa fenolik terbukti dapat menginhibisi pembentukan kristal kalsium oksalat. Oleh karena itu, tujuan penelitian ini adalah menganalisis kandungan fenolik total dan menguji aktivitas inhibisi ekstrak kulit batang Aglaia elaeagnoidea terhadap pembentukkan kristal kalsium oksalat. Proses analisis dan pengujian dimulai dengan mengekstrak serbuk kulit batang Aglaia elaeagnoidea menggunakan n-heksana, etil asetat, dan metanol menggunakan metode maserasi bertingkat. Analisis kandungan fenolik total dilakukan dengan metode kolorimetri menggunakan pereaksi Folin-Ciocalteu dengan asam galat sebagai standar fenolik yang diukur menggunakan spektrofotometer UV-Vis, dan diperoleh kandungan fenolik pada ekstrak n-heksana, etil asetat dan metanol sebesar 6,75; 14,90; dan 104,97 mg GAE/g ekstrak. Hasil pengujian aktivitas inhibisi ekstrak kulit batang Aglaia elaeagnoidea terhadap pembentukan kristal kalsium oksalat menunjukan bahwa ekstrak metanol pada kondisi lingkungan glukosa memiliki aktivitas paling tinggi dengan nilai IC50 203,17 mg/L. Proses inhibisi pembentukan kristal kalsium oksalat terjadi dengan adanya zat aktif yang teradsorpsi pada permukaan kristal dan membentuk lapisan tunggal yang berinteraksi secara fisisorpsi yang berlangsung secara spontan. Aktivitas inhibisi pembentukan kristal kalsium oksalat dipengaruhi oleh kandungan fenolik pada ekstrak kulit batang Aglaia elaeagnoidea, dengan meningkatnya kandungan fenolik pada ekstrak maka aktivitas inhibisi yang terjadi juga mengalami peningkatan. ENGLISH : Calcium oxalate is the main component in kidney stones that are formed due to the occurrence of supersaturation in the urine. Medical and traditional methods of treatment and prevention of kidney stone disease still do not provide effective results, so that efforts to find plants that have the potential to prevent kidney stone formation continue to be carried out. Aglaia elaeagnoidea plants, especially the stems, are known to have high phenolic content where phenolic compounds have been shown to inhibit the formation of calcium oxalate crystals. Therefore, the purpose of this study was to analyze the total phenolic content and to examine the inhibitory activity of Aglaia elaeagnoidea bark extract on the formation of calcium oxalate crystals. The process of analysis and testing begins with extracting the bark powder of Aglaia elaeagnoidea using n-hexane, ethyl acetate, and methanol using the multilevel maceration method. Analysis of total phenolic content was carried out by colorimetric method using Folin-Ciocalteu reagent with gallic acid as phenolic standard measured using UV-Vis spectrophotometer, and obtained phenolic content of n-hexane, ethyl acetate and methanol extracts of 6.75; 14.90; and 104.97 mg GAE/g extract. The results of the inhibitory activity of the bark extract of Aglaia elaeagnoidea on the formation of calcium oxalate crystals showed that the methanol extract under glucose environmental conditions had the highest activity with an IC50 value of 203.17 mg/L. The process of inhibition of the formation of calcium oxalate crystals occurs in the presence of the active substance which is adsorbed on the crystal surface and forms a single layer that interacts with physisorption which occurs spontaneously. The inhibitory activity of calcium oxalate crystal formation was influenced by the phenolic content in the bark extract of Aglaia elaeagnoidea, with increasing phenolic content in the extract, the inhibitory activity that occurred also increased.
