Abstract

INDONESIA: Bakteri termofilik merupakan salah satu jenis bakteri yang dihasilkan dari berbagai wilayah ekstrim, salah satunya sumber air panas. Bakteri termofilik yang dihasilkan dari sumber air panas memiliki potensi besar sebagai penghasil enzim yang stabil pada suhu tinggi. Enzim termostabil banyak digunakan dalam bidang industri untuk menunjang proses produksi. Salah satu enzim yang banyak digunakan dalam bidang industri adalah enzim protease. Tujuan dari penelitian ini adalah untuk mengidentifikasi dan mengkarakterisasi bakteri termofilik penghasil enzim protease dari sumber air panas Kawah Papandayan. Pada penelitian ini dilakukan isolasi dan identifikasi bakteri termofilik penghasil enzim protease dari sumber air panas Kawah Papandayan. Metode yang dilakukan antara lain analisis secara mikrobiologi, biokimia, penentuan kadar protein menggunakan metode Bradford, dan penentuan aktivitas protease menggunakan metode Walter yang telah dimodifikasi. Bakteri yang terisolasi memiliki karakteristik gram positif, berbentuk basil, hasil uji biokimia positif pada uji TSIA (asam), uji molilitas, dan uji MR. Isolat bakteri positif penghasil protein terbesar ditunjukan oleh BKP1 sebesar 0,0328 mg/mL, dan nilai aktivitas protease tertinggi ditunjukan oleh isolat bakteri BKP3 dengan aktivitas sebesar 0,0625 U/mL. ENGLISH : Thermophilic bacteria are one type of bacteria produced from various extreme areas, one of which is hot springs. Thermophilic bacteria produced from hot springs have great potential as producers of enzymes that are stable at high temperatures. Thermostable enzymes are widely used in the industrial field to support the production process. One enzyme that is widely used in the industrial field is protease enzyme. The purpose of this study was to identify and characterize protease enzyme-producing thermophilic bacteria from Papandayan crater hot springs. In this study, isolation and identification of thermophilic bacteria producing protease enzyme from hot springs of Papandayan crater were carried out. The methods carried out include microbiological and biochemical analysis, with the determination of protease activity using the modified Walter method. Isolated bacteria have gram-positive characteristics, bacil shaped, positive biochemical test results in the TSIA (acid) test, mollility test, and MR test. The largest protein-producing positive bacteria isolate was shown by BKP1 at 0,0328 mg/mL, and the highest protease activity value was shown by isolate BKP3 with an activity of 0.0625 U/mL.
