Abstract
INDONESIA : Tanaman Pule pandak (Rauwolfia Serpentina Benth ex. Kurz) merupakan tanaman obat potensial dari famili Apocynaceae yang mempunyai manfaat sebagai obat yang dapat menyembuhkan penyakit tekanan darah tinggi, transqulizer (penenang), dan gangguan pada sistem sirkulator. Seiring dengan bertambahnya penggunaan tanaman pule pandak sebagai obat maka keberadaannya pun akan semakin berkurang, hal tersebut dikarenakan sering dilakukan pemanenan pada bagian akarnya. Oleh karena itu, perlu dilakukan budidaya melalui kultur jaringan. Penelitian ini bertujuan untuk mengetahui pengaruh penempatan posisi eksplan daun terhadap pertumbuhan kalus pule pandak secara in vitro. Penelitian dilaksanakan di Laboratorium Kultur Jaringan Jurusan Agroteknologi, Universitas Islam Negeri Sunan Gunung Djati Bandung mulai April sampai Oktober 2016. Eksplan yang digunakan berupa daun muda tanaman pule pandak. Perlakuan yang digunakan terdiri dari 1 faktor 2 perlakuan, yaitu posisi penanaman secara abaksial dan posisi penanaman secara adaksial dan diulang sebanyak 25 ulangan. Analisis hasil penelitian dilakukan dengan metode deskriptif. Hasil penelitian menunjukkan bahwa posisi penanaman secara adaksial (posisi tulang daun diatas) mengalami pembentukan kalus yang lebih cepat, yaitu 15 HSI dengan persentase eksplan yang membentuk kalus sebesar 100%, dengan warna kalus hijau dan struktur kalus kompak. ENGLISH : Pule Pandak (Rauwolfia serpentina Benth ex. Kurz) is a potential medicinal plant from the family Apocynaceae having benefits as a medicine that cancure high blood pressure, transqulizer (tranquilizers), and disorders of the circulatory system. Due to the increasing use of Pule Pandak as medicine, its presence will be increasingly diminished because harvesting is often done on the roots. Therefore, it is necessary cultivation through tissue culture. This study aims to determine the effects of petiole placement to callus induction, and determine the best treatment on the growth of pule pandak callus in in vitro. The research conducted at Tissue Culture Laboratory of Agrotechnology Department, State Islamic University Sunan Gunung Jati Bandung from April to October 2016. The explants used young leaves of pule Pandak. Treatment consisted of one factor two treat, the position abaxial and adaxial planting position and repeated as many as 25 replications. Result analysis conducted by descriptive method. The results showed that adaxial planting position position of the veins above showed a faster callus formation, that was 15 HIS with percentase explant callus of 100%,with green callus and compact structure.
