Abstract

INDONESIA: Dalam banyak kasus penderita diabetes sering mengalami kesulitan dalam penyembuhan luka dan berujung pada amputasi. Hiperglikemia merupakan gejala awal dari penyakit diabetes yang ditandai dengan tingginya level glukosa dalam darah. Tanaman mangga memiliki manfaat sebagai tanaman obat karena mengandung senyawa metabolit sekunder yang dapat membantu dalam proses penyembuhan luka. Tujuan penelitian ini yaitu untuk mengetahui pengaruh ekstrak pucuk daun mangga terhadap level glukosa darah ikan zebra (Danio rerio) hiperglikemia akibat induksi aloksan dan glukosa serta untuk mempelajari peran ekstrak pucuk daun mangga terhadap regenerasi sirip kaudal ikan zebra. Penelitian ini menggunakan metode eksperimental dengan Rancangan Acak Lengkap (RAL) 5 perlakuan dan 10 kali ulangan. Kelompok perlakuan terdiri dari kelompok kontrol, hiperglikemia, metformin 0,75 b/v, ekstrak 10 µg/ml dan ekstrak 20 µg/ml. Ikan zebra hiperglikemia diamputasi sirip kaudalnya dan dikelompokkan ke dalam kelompok perlakuan. Parameter yang diamati meliputi level glukosa darah, pertambahan panjang sirip kaudal ikan zebra dan kesintasan. Hasil penelitian menunjukkan bahwa pemberian ekstrak pucuk daun mangga 20 µg/ml efektif dalam menurunkan level glukosa darah yaitu mencapai 68,31% dan mendekati perlakuan metformin 0,75 b/v dengan penurunan level glukosa darah mencapai 70,1%. Berdasarkan uji signifikansi menunjukkan bahwa pemberian ekstrak 20 µg/ml berbeda signifikan dengan kelompok hiperglikemia dan ekstrak 10 µg/ml. Ekstrak 20 µg/ml efektif dalam membantu proses regenerasi dengan pertambahan panjang sirip kaudal mencapai 0,094 cm selama masa perlakuan. Kesimpulannya yaitu ekstak pucuk daun mangga 20 µg/ml efektif dalam menurunkan level glukosa darah serta dapat membantu dalam proses regenerasi sirip kaudal ikan zebra. ENGLISH: In many cases, diabetics often have difficulty wound healing and lead to amputation. Hyperglycemia is an early symptom of diabetes mellitus with characterized by high levels of blood glucose. Mango plants has benefiits as a medicinal plant because it has secondary metabolite that can help in wound healing process. The purpose of this research to determine the effect of mango shoot extract on blood glucose levels of zebrafish (Danio rerio) hyperglycemia that induced by alloxan and glucose and to study the role of mango shoot extract on zebrafish caudal fin regeneration. This research was an experimental method with 5 treatment and 10 replication. The treatment group are control, hyperglycemia, metformin 0,75 b/v, extract 10 µg/ml and extract 20 µg/ml. The hyperglicemic zebrafish had its caudal fin amputated and grouped into the treatment group. The parameters observed are blood glucose levels, length of caudal fin zebrafish, and survival rate. The result showed that mango shoot extract 20 µg/ml effective for reduce blood glucose levels reaching 68,31%. It’s closer with metformin 0,75 b/v treatment with the ability reduce blood glucose levels reaching 70,1%. Based on significance test showed that extract 20 µg/ml was significantly different from the hyperglicemic group and extract 10 µg/ml. Extract 20 µg/ml was effective for regeneration process with the addition of caudal fin length reacing 0,094 cm during the treatment period. The conclusion that the extract 20 µg/ml is effective to reduce blood glucose levels and helping for regeneration on wound healing process.
