Abstract
INDONESIA : Stres merupakan pemicu timbulnya berbagai penyakit degeneratif yang disebabkan oleh faktor fisik, psikologis dan lingkungan yang tidak sehat. Stres dapat meningkatkan radikal bebas dalam tubuh dan menyebabkan stres oksidatif. Jus microgreens Pakcoy (Brassica rapa L. Chinensis) kaya akan antioksidan dan dapat dimanfaatkan dalam meredam radikal bebas. Penelitian ini bertujuan untuk mengetahui pengaruh jus microgreens Pakcoy terhadap stres pada Lalat Buah (D. melanogaster) yang diinduksi Metotreksat. Penelitian ini menggunakan metode Rancangan Acak Lengkap pada empat perlakuan dengan tiga ulangan yaitu : P0 (Kontrol), P1(MTX), P2 (jus microgreens Pakcoy) dan P3 (MTX + jus microgreens Pakcoy). Parameter yang diukur yaitu pengamatan kelulusan hidup, kemampuan memanjat, kadar katalase, dopamin, kadar klorofil, karotenoid dan kekuatan antioksidan. Hasil menunjukan bahwa paparan MTX (P1) mengakibatkan kematian terbesar D. melanogaster sebesar 24%, sedangkan penambahan jus microgreens Pakcoy (P3), kematian menjadi lebih rendah yaitu 12%. Hasil Pengamatan lokomotor yaitu pada perlakuan MTX (P1) dengan kemampuan memanjat terendah yaitu 58%, sedangkan perlakuan MTX+Jus microgreens Pakcoy (P3), kemampuan memanjat menjadi 74%. Pemberian jus microgreens Pakcoy (P2) memiliki kadar katalase tertinggi sebesar 0,049 unit/mg Protein, dan terendah pada perlakuan MTX (P1), sebesar 0,021 unit/mg Protein. Kadar dopamin tertinggi terdapat pada perlakuan jus microgreens pakcoy (P2) yaitu 0,082 ng/mg masa tubuh dan kadar terendah pada perlakuan MTX (P1) yaitu 0,027 ng/mg masa tubuh. Jus microgreens Pakcoy memiliki kadar klorofil total 3,50 mg/ml, karotenoid 33,9 mg/ml, dan kekuatan antioksidan yang kuat, jus microgreens pakcoy berpotensi dalam menekan gejala stres pada D. melanogaster. ENGLISH : Stress triggers various degenerative diseases caused by physical, psychological and environmental factors that are not healthy. Stress can increase free radicals in the body and cause oxidative stress. Microgreens Pak Choi juice is rich in antioxidants and can be used to reduce free radicals. This study aimed to determine the effect of -microgreens pak choi juice on Methotrexate-induced stress in D. melanogaster. This study used a completely randomized design method in four treatments with three replications, namely: P0 (Control), P1 (MTX), P2 (Microgreens Pak choi Juice) and P3 (MTX + microgreens pak choi juice). Parameters measured were the observation of survival rate, climbing assay, catalase, dopamine, chlorophyll, carotenoids and antioxidant power. The results showed that P1 resulted in the highest mortality of D. melanogaster (24%), while P3 resulted in lower mortality (12%). The results of locomotor observations were in the P1 with the lowest climbing assay, namely 58%. While the P3, the climbing assay became 74%. The result of P2 had the highest catalase of 0.049 units/mg protein, and the lowest was in the P1 of 0.021 units/mg protein. The highest dopamine in the P2 (0.082 ng/mg body mass) and the lowest in the P1 0.027 ng/mg body mass. Microgreens pak choi juice has total chlorophyll 3,50 mg/ml, carotenoids 33.9 mg/ml, and strong antioxidant power, microgreens pak choi juice has the potential to suppress stress symptoms in D. melanogaster.
