Abstract
INDONESIA : α-Amilase merupakan enzim dengan permintaan yang cukup tinggi dalam berbagai proses industri makanan. α-Amilase memiliki kemampuan menghidrolisis ikatan α-1,4 glikosida pada bagian struktur pati sehingga dapat menghasilkan disakarida dan monosakarida yang berukuran lebih sederhana. Penelitian ini bertujuan untuk isolasi ekstrak kasar α-amilase dan menentukan aktivitas α-amilase dari bakteri termofilik yang berasal dari sumber pemandian air panas Way Belerang desa Kecapi kecamatan Kalianda kabupaten Lampung menggunakan metode DNS dan konsentrasi protein dengan metode Bradford sehingga diperoleh aktivitas spesifik. Pada penelitian ini dilakukan 2 tahapan yaitu isolasi α-amilase dan fraksinasi dengan penambahan ammonium sulfat pada tingkat kejenuhan 0-20%, 20-40% dan 40-60% selanjutnya dilakukan dialisis untuk menghilangkan garam dan zat pengotor hasil fraksinasi. Hasil penelitian menunjukan aktivitas α-amilase tertinggi pada fraksi 20-40% yaitu sebesar 269,88 U/ml dan konsentrasi protein 10,313 mg/L sehingga diperoleh aktivitas spesifik sebesar 26201,9417 U/mg. ENGLISH: α-Amylases are enzymes with high demand in various food industry processes. α-Amylases have the ability to hydrolyze α-1,4 glycoside bonds in the starch structure so that they can pr oduce disaccharides and monosaccharides that are simpler in size. This study aimed to isolate crude α-amylase extract and determine the α-amylase activity of thermophilic bacteria originating from the Way Sulfur Hot Spring, Kecapi Village, Kalianda District, Lampung Regency using the DNS method and protein concentration with the Bradford method in order to obtain specific activity. In this study, two stages were carried out, namely isolation of α-amylase and fractionation with the addition of ammonium sulfate at saturation levels of 0-20%, 20-40% and 40-60%, then dialysis was carried out to remove salt and impurities from the fractionation. The results showed the highest -amylase activity in the 20-40% fraction, which was 269,88 U/ml and the protein concentration was 10,313 mg/L so that the specific activity was 26201,9417 U/mg.
