Abstract
INDONESIA: Stroberi (Fragaria sp.) merupakan tanaman subtropis yang saat ini sudah dibudidayakan di Indonesia. Kebutuhan para petani akan bibit bebas penyakit dan unggul masih kurang. Karena perbanyakan bibit dengan stolon masih memiliki kekurangan. Maka itu digunakan alternatif perbanyakan pembibitan stroberi antara lain adalah dengan kultur jaringan yang akan mendapatkan bibit stroberi yang bebas penyakit dan unggul. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh serta menentukan konsentrasi optimum NAA terhadap induksi kalus Fragaria sp. var Sweet Charlie. Penelitian dilakukan di Laboratorium Kultur Jaringan UIN Sunan Gunung Djati Bandung pada bulan Januari-Mei 2022. Penelitian ini merupakan penelitian eksperimental menggunakan Rancangan Acak Lengkap (RAL) dengan 5 taraf yaitu (NAA 0 ppm, NAA 0,5 ppm, NAA 1 ppm, NAA 1,5 ppm, dan NAA 2 ppm) dengan 5 perlakuan masing-masing 3 kali pengulangan. Hasil penelitian menunjukkan konsentrasi 1,5 ppm NAA efektif terhadap waktu muncul kalus 12 HST, warna kalus terbaik adalah warna putih terdapat pada konsentrasi 1,5 ppm dengan tekstur remah. Konsentrasi 1,5 ppm NAA efektif terhadap diameter kalus sebesar 2,1 cm, persentase pembentukan kalus tertinggi terdapat pada konsentrasi 1,5 ppm sebesar 100%. Adanya pengaruh yang diberikan NAA terhadap pertumbuhan kalus dengan konsentrasi optimum pada 1,5 ppm. ENGLISH: Strawberry (Fragaria sp.) is a subtropical plant which is currently cultivated in Indonesia. The need for farmers for disease-free and superior seeds is still lacking. Because seed propagation with stolons still has drawbacks. Therefore, an alternative to propagation of strawberry nurseries is used, including tissue culture which will produce disease-free and superior strawberry seedlings. The purpose of this study was to determine the effect and determine the optimum concentration of NAA on callus induction of Fragaria sp. var Sweet Charlie. The study was conducted at the Tissue Culture Laboratory of UIN Sunan Gunung Djati Bandung in January-May 2022. This research was an experimental study using a Completely Randomized Design (CRD) with 5 levels, namely (0 ppm NAA, 0.5 ppm NAA, 1 ppm NAA, 1 ppm NAA 1.5 ppm, and NAA 2 ppm) with 5 treatments each with 3 repetitions. The results showed that the concentration of 1.5 ppm NAA was effective against the appearance of callus 12 HST, the best callus color was white at a concentration of 1.5 ppm with a crumb texture. The concentration of 1.5 ppm NAA was effective against a callus diameter of 2.1 cm, the highest percentage of callus formation was found at a concentration of 1.5 ppm of 100%. There was an influence given by NAA on callus growth with an optimum concentration of 1.5 ppm.
